چکیده

We investigated the acute metabolic effects of isoflavones from Pueraria lobata (Willd.) Ohwi (IPL) in ovariectomized (OVX) mice. After 4 weeks of IPL feeding at 500 mg/day/kg body weight (OVX500), plasma 17β-estradiol concentrations were significantly higher (+25%, p < 0.05), whereas plasma triglyceride levels were significantly lower in OVX mice (-15%, p < 0.05) compared with controls. Abdominal adipose tissue weight was marginally reduced in IPL-fed groups compared with OVX controls and the plasma levels of liver enzymes were unchanged. In addition, IPL significantly inhibited the reduction of bone mineral density in the femurs of OVX mice (OVX200, +22%; OVX500, +26%; p < 0.05) compared with controls after 4 weeks of IPL feeding. In quantitative polymerase chain reaction analysis the expression of aromatase was significantly suppressed and SULT1E1 was increased by IPL feeding, showing that IPL feeding may not alter the risk for breast cancer in mice. Our results suggest that IPL could ameliorate menopausal symptoms in mice. Further studies will confirm the effects of IPL in humans.

Effects of Dietary Isoflavones from Puerariae Radix on Lipid and Bone Metabolism in Ovariectomized Rats

Puerariae radix, the dried root of Pueraria lobata Ohwi, is one of the earliest and most important edible crude herbs used for various medical purposes in oriental medicine. This study evaluated the metabolic effects of total isoflavones from P. lobata (PTIF) in ovariectomized (OVX) rats. The OVX rats were divided into four groups treated with distilled water, 17β-estradiol (E2 10 μg/kg, once d...

Studies on the extraction process of total flavonoids in Radix puerariae and their hypoglycemic effect in mice

Radix Puerariae is the dried root of Pueraria lobata (Willd.) Ohwi, which releases a variety of isoflavones. The objective is to investigate the extraction process of total flavonoids in Radix Puerariae and to examine their effects on blood glucose MDA in alloxan-diabetic mice. Solvent extraction and column chromatographic isolation were used to extract the total flavonoids in Radix Puerariae, ...

Long-Term Treatment of Puerariae Radix Extract Ameliorated Hyperparathyroidism Induced by Ovariectomy in Mature Female Rats

Postmenopausal osteoporosis is a disorder characterized by the progressive bone loss induced by estrogen deficiency in postmenopausal women. This imbalance affects calcium–phosphate metabolism and results in secondary hyperparathyroidism. Purariae Radix (PR), the root of P. lobata (Wild.) Ohwi, is one of the earliest medicinal herbs employed in ancient China. PR contains a high quantity of isof...

Aged people, especially postmenopausal women are at increased risk of metabolic disorders such as obesity, which in turn raises the risk of developing further serious conditions, including diabetes [1,2], cancers, neurological disorders, hypertension, and other cardiovascular diseases [3]. Postmenopausal diabetes is induced by estrogen deficiency, which exacerbates insulin resistance [4,5]. The...
